DNA Precipitation: Concentrating Nucleic Acids

To concentrate DNA, precipitation is performed using ice-cold ethanol or isopropanol in the presence of salts (e.g., sodium acetate). The alcohol reduces DNA solubility, causing it to form visible white filaments or a pellet upon centrifugation. This step also helps remove residual salts and solvents. The precipitated DNA is collected by centrifugation and washed with 70% ethanol to further purify and desalt it. Precipitation ensures the DNA is concentrated into a manageable volume for subsequent handling.

Washing and Resuspension: Obtaining Pure DNA

The final step involves washing the DNA pellet to remove any remaining impurities and resuspending it in a suitable buffer. The pellet is washed with 70% ethanol to remove traces of salt and alcohol, then air-dried to evaporate residual ethanol. Finally, the DNA is dissolved in TE buffer or nuclease-free water. The buffer stabilizes the DNA and prevents degradation, ensuring it is ready for use in PCR, sequencing, restriction digestion, and other molecular biology applications.
